Molecular Recognition within foldamers

Molecular recognition relies on the convergence of an array of chemical functions towards a binding pocket. Nature’s design relies on the self-organization of oligomeric or polymeric sequences offering unmatched modularity in that each and every individual monomer may be varied in order to tune the structure, the dynamics and host-guest properties of the folded conformation. Using foldamers, complex artificial architectures based on long sequences of monomers are now easily amenable to re-combination and evolution. In essence, this research amounts to creating new, artificial, codes between primary one-dimensional sequences of aromatic monomers, tertiary three-dimensional folded structures of oligomers, and molecular recognition properties to develop fully artificial foldamer-based receptors.

Although bio-inspired, artificial foldamer sequences differ from natural backbones and are expected to give rise to unique structures and properties. While biopolymer diversity arises from variations of the sequence of side-chains, diversity in foldamer sequences can also stem from unlimited variations of the main chain components inasmuch as they possess compatible folding patterns. We and others have proposed a unique design of helically folded capsules based on sequences of aromatic amino-acids which code for a large helix hollow in the center and a narrow helix diameter at the end of the sequence.

A key challenge in the foldamer area is to fully exploit the immense variety of monomers available in order to identify the best sequence candidates to act as selective receptors. Using a vast library of aromatic amino acid monomers available in our lab, we developed a structure-based iterative evolution strategy which allows the preparation of helically folded aromatic oligoamide sequences for the selective encapsulation of a given guest
